WHAT IS KISSPEPTIN-10?

Kisspeptin-10 is a decapeptide — the shortest bioactive fragment of the kisspeptin family, derived from the KISS1 gene product that also yields the longer kisspeptin-54, -14, and -13 fragments. It is described in the literature as the minimal kisspeptin sequence with full intrinsic bioactivity, and is studied within the hypothalamic-pituitary-gonadal (HPG) axis — the signalling system central to puberty and reproductive endocrinology. This guide summarises the published literature on kisspeptin-10 for laboratory researchers.

RESEARCH BACKGROUND

  • HPG axis regulation, puberty onset, and reproductive hormone signalling — with strong human genetic evidence: loss-of-function mutations in the kisspeptin receptor cause hypogonadotropic hypogonadism, while gain-of-function mutations cause precocious puberty
  • Human pharmacology: a randomised infusion study in healthy men found continuous low-dose kisspeptin administration increased LH pulse frequency, secretory burst mass, FSH, and testosterone
  • Hypothalamic amenorrhea research, where kisspeptin signalling has been studied as a way to stimulate GnRH neuron responsiveness
  • IVF and reproductive medicine research, where the longer-acting kisspeptin-54 fragment (not kisspeptin-10 itself) has been studied as an ovulation trigger with a reduced ovarian hyperstimulation syndrome (OHSS) signal compared to conventional triggers

A precision note for researchers: much of the strongest human clinical trial work in this space (IVF trigger studies, hypothalamic amenorrhea research) used the longer kisspeptin-54 fragment rather than kisspeptin-10 specifically, due to KP-10’s very short half-life (below). The two share the same receptor mechanism but differ in duration of action and the specific studies that have used them.

PROPOSED MECHANISM OF ACTION

Kisspeptin-10 is an agonist at KISS1R (also known as GPR54), a G-protein-coupled receptor expressed predominantly on hypothalamic GnRH neurons. Receptor activation triggers GnRH release, which cascades to pituitary luteinising hormone (LH) and follicle-stimulating hormone (FSH) secretion. This pathway is one of the most genetically well-validated in reproductive endocrinology, given the human mutation data described above.
