SM1 (Semaglutide)
C₁₈₄H₂₉₁N₄₅O₅₉ · GLP-1 Receptor Agonist · MW 4,113.6 Da
This SM1 research guide covers its structure, mechanism, research applications and available sizes — the foundational GLP-1 receptor agonist research compound, extensively characterised and the standard reference for GLP-1 pathway research globally.
What Is SM1?
SM1 is a synthetic analogue of native human GLP-1 (Glucagon-Like Peptide-1), sharing approximately 94% amino acid sequence homology with the endogenous hormone. It incorporates a C18 fatty diacid modification attached via a hydrophilic linker at position 26, enabling strong albumin binding and dramatically extending its half-life compared to native GLP-1.
Native GLP-1 has a half-life of only 1–2 minutes due to rapid degradation by the enzyme DPP-4 (dipeptidyl peptidase-4), as documented in peer-reviewed pharmacology literature. Its structural modifications — including a substitution at position 8 that confers DPP-4 resistance — extend the compound's half-life to approximately 7 days in relevant models, making it far more practical for extended laboratory protocols.
SM1 is the most extensively published GLP-1 receptor agonist compound in the scientific literature, making it the essential reference compound for any GLP-1 pathway research programme. It is available from Pyrox Labs in 5mg and 10mg individual research vials.
Research Use Only: SM1 is supplied by Pyrox Labs exclusively for in vitro laboratory research and independent scientific study. Not for human consumption, self-administration or therapeutic use.
Structural Modifications
Three key structural modifications distinguish it from native GLP-1 and contribute to its extended half-life and receptor binding characteristics:
GLP-1 Receptor Pathway
It acts as a full agonist at the GLP-1 receptor (GLP-1R), a class B G protein-coupled receptor. GLP-1R activation triggers cyclic AMP (cAMP) signalling cascades with downstream effects in multiple tissues. Key research areas include pancreatic beta cell signalling, gastric motility modulation, hypothalamic appetite-regulating pathways and cardiovascular function in relevant research models. Downstream ERK1/2 and PKA signalling cascades are also commonly used as functional readouts in receptor activation assays.

Sizes & Pricing
This SM1 research guide includes full sizing and reconstitution details below. It's available in individual research vials and as part of a 20mg research bundle. All pricing subject to confirmation.
Storage & Reconstitution
Store the lyophilised compound at room temperature away from direct light and moisture. Once reconstituted with bacteriostatic water, store refrigerated at 2–8°C and use within 28 days. Do not freeze reconstituted solution. It reconstitutes clearly — any cloudiness or particulate matter after thorough swirling may indicate degradation.
