WHAT IS KISSPEPTIN-10?
Kisspeptin-10 is a decapeptide — the shortest bioactive fragment of the kisspeptin family, derived from the KISS1 gene product that also yields the longer kisspeptin-54, -14, and -13 fragments. A precision note for researchers: much of the strongest human clinical trial work in this space (IVF trigger studies, hypothalamic amenorrhea research) used the longer kisspeptin-54 fragment rather than kisspeptin-10 specifically, due to KP-10’s very short half-life (below). The two share the same receptor mechanism but differ in duration of action and the specific studies that have used them.
PROPOSED MECHANISM OF ACTION
Kisspeptin-10 is an agonist at KISS1R (also known as GPR54), a G-protein-coupled receptor expressed predominantly on hypothalamic GnRH neurons. Receptor activation triggers GnRH release, which cascades to pituitary luteinising hormone (LH) and follicle-stimulating hormone (FSH) secretion. This pathway is one of the most genetically well-validated in reproductive endocrinology, given the human mutation data described above.
