WHAT IS DSIP?
DSIP (Delta Sleep-Inducing Peptide) is a nonapeptide originally isolated in 1974 from the cerebral venous blood of rabbits in induced slow-wave sleep, by the Schoenenberger–Monnier group in Switzerland. It was later studied by both Western European and Soviet/Russian research groups. This guide summarises the published literature on DSIP for laboratory researchers — including an important scientific caveat that researchers should be aware of before working with it.
RESEARCH BACKGROUND

- Early human studies (small samples, 1980s) reporting effects on sleep duration, interruption frequency, and REM sleep following intravenous administration
- HPA-axis research, including a study finding DSIP reduced CRF-induced corticosterone release in preclinical models
- Broader physiological effects reported through the 1980s beyond sleep, including stress and withdrawal-related research
An important scientific caveat: the definitive modern review of this compound — Kovalzon & Strekalova, Journal of Neurochemistry, 2006, titled “Delta sleep-inducing peptide (DSIP): a still unresolved riddle” — explicitly states that DSIP’s existence as a discrete, well-defined endogenous regulatory peptide has not been robustly established. The original sleep-inducing effect reported in the 1970s work has not been reliably replicated in later, more rigorous research, and effects attributed to “DSIP” may reflect DSIP-like immunoreactive material bound to carrier proteins rather than a single free bioactive peptide with a defined receptor. There is essentially no active modern (post-2010) research programme investigating DSIP pharmacology — the published literature is dominated by primary work from the 1970s–1980s and periodic critical reviews. PROPOSED MECHANISM OF ACTION
No specific DSIP receptor has ever been isolated or cloned, and its precursor protein was never conclusively identified. Proposed (speculative) mechanisms in the literature include interaction with the MAPK/ERK signalling cascade and structural homology to the glucocorticoid-induced leucine zipper (GILZ) protein. Given the unresolved questions raised by the modern literature review cited above, researchers should treat any described mechanism for DSIP as a historical hypothesis rather than settled pharmacology.
