WHAT IS SEMAX?
Semax is a synthetic heptapeptide, an analogue of ACTH(4โ10) โ a fragment of adrenocorticotropic hormone โ with an added Pro-Gly-Pro stabilising motif. Despite its ACTH-derived structure, published research indicates Semax does not carry the hormonal/adrenocortical activity of full-length ACTH. Developed in the USSR/Russia in the 1980s, Semax has more regulatory history than most compounds in this Knowledge Hub โ it has been registered in Russia as a nasal-drop pharmaceutical for a specific indication. RESEARCH BACKGROUND

- Neurotrophin signalling: rodent studies found a single dose of Semax increased hippocampal BDNF protein and trkB receptor phosphorylation, alongside increased BDNF gene transcription
- Stroke recovery research: a published human clinical study (30 acute ischaemic stroke patients vs. 80 conventional-therapy controls) reported improved regression of focal neurological deficits; a related study examined Semax in chronic cerebrovascular insufficiency
- Rodent stress and memory research, where Semax was found to attenuate stress-induced impairment of memory-task performance
- Dopaminergic and serotonergic system modulation, studied across several rodent brain regions
A note on the evidence base: the human stroke studies are real and PubMed-indexed, and represent some of the more clinically substantial data among the compounds in this guide series โ but they are dated (late 1990sโ2000s), conducted in Russian patient populations, and have not been replicated in Western randomised controlled trials. As with Selank, essentially all foundational Semax pharmacology traces back to a small number of Russian research groups.
PROPOSED MECHANISM OF ACTION
Semax is studied for its effects on neurotrophin signalling โ specifically upregulation of BDNF (brain-derived neurotrophic factor) and NGF (nerve growth factor) expression in the hippocampus, frontal cortex, and retina, with downstream trkB receptor activation. A melanocortin receptor interaction (MC4R has been proposed) has also been suggested as a possible binding mechanism, though this is less rigorously characterised than for classical melanocortin agonists. Semax is also studied for inhibition of enkephalin-degrading enzymes, a mechanism shared conceptually with Selank, and has been shown in rodent studies to lack the HPA-axis/cortisol activity associated with full-length ACTH.

Is Semax approved for human use?
The Semax sold by Pyrox Labs is not approved by the FDA, MHRA, or any UK/EU/US regulatory body for human or veterinary use. Semax does have a separate pharmaceutical registration history in Russia; that registration does not apply to the research-grade compound supplied by Pyrox Labs.
Can Pyrox Labs advise on dosing or administration?

Is Semax the same family of peptide as Selank?
